Abstract:

For breeding early-ripen nectarine varieties in order to avoid fruit fly damage in Taiwan lowland area began from May. This program chose peach and nectarine varieties which had short fruit development period and low chilling requirement as female parent and used embryo rescue technique to culture immature hybrid embryos. Embryo aborted when ‘Armking’ and ‘Armking Precoce’ fruits became soft. The optimum time for culturing embryos was when fruits were firm ripe. The best results came from embryos were cultured on Murashige and Skoog medium better than those on Nitsch and Nitsch or on Steward and Hsu medium. Supplemented with 1 mg/l BA + 1 mg/I IAA or 1 mg/BA + 1 mg/I IBA on MS basal medium was better than supplement with any single plant growth regulators or combinations. However, no significant effect between MS medium with or without plant growth regulators. Embryos germinate difficulty without 4°C treatment and usually produced plants that rosette and are difficult to grow. Transplanting the growing embryo from the culture tube to soil is the most critical stages for successful embryo culture in this program.
