Abstract:
Shih-hu is an important traditional Chinese medicine. Due to some of shih-hu were near extinct for protection, an artificial seedling proliferation system for shih-hu is required urgently. An efficient protocol for in vitro propagation and development of D. moniliforme (L.) Sweet using the germinated seedling has been developed. The germinated seedlings (1 cm height) were sub-cultured on half-strength Murashige and Skoog’s (MS) basal medium supplemented with different vegetable and fruit homogenate in various concentrations and 3% sucrose for further growth. The optimum fresh weight of seedlings after three months culturing were found on 80~100 g/l banana homogenate treatments. The longest height of plantlet were found on 80 g/l yambean homogenate. An increase in dry/fresh weight index was appeared when subculture seedling on the medium supplemented with 80~100 g/l potato homogenate which might be beneficial for acclimation in vitro seedling.
