Ⅲ. Effect of Cefotaxime and Plant Growth Regulators on Growth of Shoot Tip Explants in Vitro

Abstract:
This paper describes a method of in vitro propagation of Heliconia psittacorum cv. Rhizomatosa by using terminal shoot tip explants. Various concentrations (25, 50, 100 and 500 mg/l) of cefotaxime were used in liquid Murashige and Skoog (MS) basal medium to control contamination. Lower rate of contamination was observed in shoot tip explants after pretreatment of explants in liquid MS medium supplemented with 500 mg/l cefotaxime for 3 days. Well developed shoots were successfully obtained after 45 days of culture of shoot tip explants on MS medium supplemented with 4-8 mg/l Benzylaminopurine (BA) and 0.5 mg/l α - Naphthaleneacetic acid (NAA). Addition of 0.01-0.5 mg/l Thidiazuron (TDZ) in the medium was found to be beneficial for rapid growth of shoot tip explants. The effect of various culture methods on growth of shoot tip explants was also studied.
